番茄温敏核不育系花药培养诱导愈伤组织的研究 被引量:3

摘要 [目的]探讨影响番茄温敏核不育系花药培养诱导愈伤组织的相关因子。[方法]研究不同灭菌处理、不同基本培养基、不同激素种类及浓度对番茄温敏核不育系花药培养诱导愈伤组织的影响。[结果]0.1%HgCl28 min+10%NaClO8 min的灭菌处理效果最佳,菌污染率仅为1.95%,出愈率达71.73%;在MS、B5、Miller3种基本培养基的比较试验中,以MS为基本培养基对花药愈伤组织的诱导效果最好;诱导愈伤组织的适宜浓度为2,4-D1 mg/L、6-BA2 mg/L、KT2 mg/L。[结论]灭菌处理、基本培养基、激素种类及浓度可以影响番茄温敏核不育系花药培养诱导愈伤组织的效果。 [ Objective ] The aim of the study was to investigate the interrelated factors affecting the callus induction by anther culture of thermo-sensitive genie male sterile line of tomato. [ Method] This research studied the effects of different sterilization treatments, basic mediums, hormone kinds and concentrations on the anther culture for inducting callus of thermo-sensitive genie male sterile line of tomato. [ Result ] 0. 1% HgCI2 8 min + 10% NaClO 8 min was the best sterilization treatment. The contamination rate was only 1.95% and the callus induction rate achieved to 71.73%. In the comparing experiments of three kinds of basic mediums named MS, Bs and Miller, MS basic mediums had got the best effect on callus induction of the anther. The suitable concentrations of three kinds of the hormone using separately on the callus induction was 2,4-D 1 mg/L, 6-BA 2 mg/L or KT 2 mg/L. [ Conclusion] The callus induction effects on the anther culture of thermo-sensitive genie male sterile line of tomato could be affected by the sterilization treatments, basic mediums, hormone kinds and concentrations.
