广东虫草多糖脱蛋白及凝胶渗透色谱法表征其分子量分布的研究 被引量:12

摘要 通过比较蛋白脱除率和多糖损失率,研究Savage法、三氯乙酸(TCA)法及盐酸法对广东虫草多糖(CGP)脱蛋白的效果,并利用凝胶渗透色谱法(GPC)测定虫草多糖组分的保留时间tR、重均分子量Mw、数均分子量Mn、峰位分子量Mp、多分散性PD以及各组分的相对含量,以表征广东虫草多糖的分子量分布。结果表明:采用Savage法脱蛋白总体效果较好,处理次数3次为宜;GPC谱图分析结果表明,CGP含有三个不同分子量级别的多糖组分,其重均分子量Mw分别为2.57×106、3.84×104及5.00×103。 Deproteinization effect of Sevage method, trichloroacetic acid (TCA) method and hydrochloric acid (HCI) method on Cordyceps guangdongensis polysaccharide (CGP) were studied by comparison of CGP loss rate and the deprote'mization rate. In order to characterize the molecular weight distribution of CGP, retention time (tp.),weight average molecular weight (Mw) ,number average molecular weight (Mn), peak position molecular weight (Mp),polydispersity (PD) and relative content of CGP fractions were obtained by gel permeation chromatography(GPC) .The results showed that, Sevage method showed the best deproteinization effect, with which three-times processing of the CGP was suggested. GPC analysis showed that there were three constituents with different molecular weight level in CGP, the corresponding weight average molecular weight (Mw) of the three constituents were 22.57×10^6、3.84×10^4 and 5.00×10^3, respectively.
