摘要
目的探讨龈沟液中基质金属蛋白酶-2(MMP-2)含量与正畸牙根吸收的相关性,为牙根吸收的临床诊断寻找简单、特异性的检测指标。方法选取雄性SD大鼠30只,加力将上颌第一磨牙向近中移动,建立牙根吸收和牙根未吸收的正畸牙模型,每周提取实验牙的龈沟液。应用组织形态学染色观察实验大鼠的牙根吸收程度,并通过SDS-PAGE、Western Blot和JD-801凝胶成像系统定量检测各组龈沟液中MMP-2含量的改变。结果实验牙受力1周后,出现牙根吸收大鼠的龈沟液中MMP-2含量显著升高,而未出现牙根吸收大鼠的龈沟液中MMP-2水平无明显变化。牙根吸收组和未吸收组龈沟液中MMP-2含量有统计学差异(P<0.05)。结论龈沟液中的MMP-2的表达水平与正畸源性出现牙根吸收密切相关。
Objective To investigate the relationship between matrix metalloproteinase2 in gingival crevicular fluid with orthodontic root resorption, so as to find a new simple and specific method to detect the development of root resorption. Methods 30 male adult SD rats were selected to establish animal root resorption and non- root resorption models by applying an excess force to the maxillary first molar. The gingival crevicular fluids were collected every week after the start of force application. Root adsorption were evaluated by histomorphologic staining, the level of MMP-2 in gingival crevicular was assayed by biochemistry techniques of SDS-PAGE, Western Blot and JD-801 gelatum analysis system. Results 1 week after the force were applied, the level of MMP 2 in gingival crevicular fluid increased significantly in root resorption group, no significant change of level of MMP-2 in gingival crevicular fluid was discovered in non-root resorption group. The level of MMP-2 in gingival crevicular fluid was significantly different between the root resorption group and non-root resorption group (P〈0.05). Conclusion The level of MMP-2 in gingival crevicular fluid may have intimate relationship with root resorption under orthodontic force.
