摘要
为了能利用ISSR技术在分子水平上研究贵州天冬种质资源的遗传多样性,对天冬叶片DNA的提取方法进行了研究。结果表明:提取液中使用CTAB比SDS所得DNA质量好。研磨时加氯仿可以有效抑制DNA酶的活性和去除蛋白质杂质;用氯仿/异戊醇(24/1)抽提1次可获得质量好、浓度高的DNA,可以满足ISSR分析的需要。
Four DNA extraction methods from Asparagus cochinchinensis leaves was tested to study the genetic diversity of Asparagus cochinchinensis germplasm resources by using ISSR technique on the molecular level. The results showed that the DNA quality from the extraction solution with 1.5% CTAB is better than that from the extraction solution with 1.5% SDS, adding chloroform can effectively inhibit the activity of DNase and remove the impurity of protein when the sample was grinded, and the DNA with good quality and high concentration can be extracted once by chloroform/isopentanol (24/1) analysis, which can meet the needs of ISSR analysis.
