摘要 目的探讨槲皮素对结肠癌细胞系RKO中P53蛋白以及其下游基因Gadd45a表达的影响。方法采用免疫细胞化学的方法检测不同浓度槲皮素作用于RKO细胞后的P53蛋白表达的变化,采用RT-PCR与Western Blot技术检测不同浓度槲皮素作用于RKO细胞后的Gadd45amRNA和蛋白表达的变化。此外,采用Western Blot技术检测不同浓度槲皮素作用于EC109细胞后的Gadd45a蛋白表达的变化。结果槲皮素作用于RKO细胞后,细胞的P53表达增高,Gadd45amRNA和蛋白表达水平逐渐增高,并且呈现出一定浓度依赖性。另外,槲皮素作用于EC109细胞后,Gadd45a蛋白表达水平逐渐增高,也呈现出一定浓度依赖性。结论槲皮素可能通过上调P53依赖途径中p53基因表达使Gadd45a表达增高,由此介入了对肿瘤发生发展的抑制。 Objective To explore the influence of quercetin on the expression of P53 and its downstream gene, Gadd45a, in colon cancer cell hne RKO and esophageal cancer cell line EC109. Methods The expression of P53 in RKO cells was detected by immunocytochemistry, and the mRNA and protein expression of Gadd45a in RKO cell were detected by RT-PCR and Western blot, respectively. In addition,the protein expression of Gadd45a in EC109 cells was detected by Western blot. Results The expression of P53 was induced by quercetin in a dose-dependent way. Furthermore, the mRNA and protein expression of Gadd45a was also induced by quercetin in a dose-dependent way in RKO cells. The protein expression of Gadd45a also increased in a dose-dependent manner in EC109 cell line after administration of quercetin. Conclusion Quercetin could have a suppressive effect on the tumor by inducing the expression of P53 and Gadd45a.
