摘要 目的探讨CRP对血管内皮细胞泡沫化及IL-6、MCP-1表达的影响。方法不同浓度的CRP(0mg/L、5mg/L、10mg/L、20mg/L、40mg/L)与50mg/Lox-LDL同时作用于人脐静脉内皮细胞,体外培养24h后,采用油红O染色法观察细胞泡沫化情况;采用ELISA方法测定细胞培养上清液中IL-6及MCP-1的浓度。结果①随着CRP浓度的增加,人脐静脉内皮细胞内脂滴数量逐渐增多,脂滴逐渐增大,细胞泡沫化程度逐渐加重;②不同浓度CRP(5mg/L、10mg/L、20mg/L、40mg/L)和50mg/Lox-LDL共同刺激24h后,人脐静脉内皮细胞产生IL-6浓度分别为(33.90±2.64)mg/L、(40.77±3.62)mg/L、(42.19±3.82)mg/L和(44.16±0.57)mg/L,与单独50mg/Lox-LDL培养的对照组(14.41mg/L±3.38mg/L)比较,有显著性差异(P<0.05);③不同浓度CRP(5mg/L、10mg/L、20mg/L、40mg/L)和50mg/Lox-LDL共同刺激后,人脐静脉内皮细胞产生MCP-1浓度分别为(23.82±3.44)μg/L、(20.11±4.10)μg/L、(19.25±3.67)μg/L、(24.24±3.34)μg/L,与单独50mg/Lox-LDL培养的对照组(20.36μg/L±5.40μg/L)比较,无显著性差异(P>0.05)。结论高脂环境下,CRP可加重血管内皮细胞泡沫化,并通过诱导内皮细胞合成IL-6而促进动脉粥样硬化的发生与发展。 Objective To investigate the foamed cells formation and the expression of interleukin - 6 ( IL - 6) and monocyte ehe- moattractant protein - 1 ( MCP - 1 ) in human vascular endothelial cell induced by C - reactive protein (CRP). Methods Human umbili- cus vein endothelial ceils were co -incubated 24h with 50mg/L ox -LDL and different diluted concentrations of CRP (0mg/L, 5mg/L, 10mg/L, 20mg/L, and 40mg/L, respectively). Those cells were stainned by oil red and the status of the foamed endothelial cells was examed by upside- down microscope. Expression of IL-6 and MCP -1 was measured by enzyme linked immunosorbent assay (ELISA). Results ①The numbers of lipid droplet in vascular endothelial cells became more and larger as the density of the CRP increased. ②The expression of IL -6 in vascular endothelial cells induced by CRP in different diluted concentrations (5 mg/L, 10rag/L, 20mg/L, and 40rag/L, respectively) were 33.90 ± 2.64, 40.77 ± 3.62, 42.19 ± 3.82, and 44.16 ± 0.57mg/L, respectively. And those data was signifancant increased than that of in 0mg/L CRP control group ( 14.41 ± 3.38rag/L) ( P 〈 0.05 ). ③The synthesis of MCP - 1 stimulated by 0mg/L, 5mg/L, 10rag/L, 20mg/L, 40mg/L CRP in vascular endothelial cells were no statistical difference [ (20.36 ± 5.40) , (23. 82 ±3.44) ,(20.11 ±4.10),(19.25 ±3.67),(24.24±3.34)μg/L,respectively)] (P〉0.05).Conclusion CRP could accelerate the progress of foam cell formation in human vascular endothelial cells and might be through the pathway of inducing overexpression of IL -6, but not MCP - 1.
