HSA/IL1ra融合蛋白的纯化及生物学活性研究

摘要 目的:纯化HSA/IL1ra融合蛋白并对其生物学活性进行检测。方法:采用亲和层析及离子交换层析分离纯化HSA/IL1ra融合蛋白,检测其对IL1引起的对A375 S2的杀伤保护作用。结果:获得的HSA/IL1ra融合蛋白HPLC纯度在98%以上,并且具有与天然IL1ra相似的、对IL1杀伤A375 S2细胞的保护作用。结论:获得高纯度的HSA/IL1ra融合蛋白并且具有相应的生物学活性。 Objective: To purify the recombinant human serum albumin fusion protein with interleukin 1 (HAS/IL1ra) and to detect the bioactivity of the fusion protein. Methods: The recombinant HAS/IL1ra protein was purified by affinity chromatography and ion exchange chromatography,the bioactivity of the fusion protein was detected by IL1-induced A375 S2 cell killing. Results: The purity of the fusion protein was at least 98% as assessed by HPLC and the protective effect from the IL1-induced A375 S2 cell killing was similar to natural IL1ra. Conclusion: The purified recombinant HAS/IL1ra protein in this study has a satisfactory bioactivity.
