摘要
目的:探索RNA干扰(RNAi)hTR基因对膀胱尿路上皮癌BIU-87细胞株端粒酶活性的影响,为膀胱尿路上皮癌基因治疗提供新的途径及依据。方法:构建以hTR基因为靶点的RNA干扰装置PhTR-siRNA,对其进行筛选和鉴定;检测PhTR-siRNA对BIU-87细胞的生物学效应,包括抑制BIU-87细胞的生长情况、端粒酶活性检测、实时定量PCR(RealTime-PCR)验证hTR变化。结果:PhTR-siRNA干扰hTR基因后膀胱尿路上皮癌BIU-87细胞株的生长受到抑制,明显抑制端粒酶活性,实时定量PCR验证hTR基因明显减少。结论:RNA干扰hTR基因能明显抑制膀胱尿路上皮癌BIU-87细胞株的生长,可作为一种膀胱尿路上皮癌的基因治疗方法。
To investigate the influence of RNA interference of the hTR gene (RNAi-hTR) on the telomerase activity of the bladder cancer BIU-87 cell line. Methods : We constructed, screened and identified the RNAi system-PhTR-siRNA targeting hTR, analyzed the biological effect of PhTR-siRNA on BIU-87 cells, including its inhibition of their growth, determined telomerase activity and detected the changes of the hTRgene by RT-PCR. Results: RNAi-hTRobviously inhibited the growth of bladder cancer BIU-87 cells, suppressed telomerase activity and markedly decreased the hTR gene. Conclu- sion : RNAi-hTR can considerably inhibit the growth of bladder cancer BIU-87 cells. It may be one of the most promising gene therapeutic strategies for bladder cancer.
