缺氧低糖培养对少突胶质前体细胞的影响 被引量:2

摘要 目的:探讨缺氧低糖环境对体外培养少突胶质前体细胞(O2A细胞)的影响。方法:应用台盼蓝染色计数细胞存活率,Hochest33258荧光染色及流式细胞仪分析细胞凋亡情况,透射电镜观察细胞形态变化。结果:缺氧培养3h细胞变化不明显,但缺氧培养6h,细胞存活率明显下降,凋亡细胞明显增多,并且随着缺氧时间延长存活细胞更少、凋亡细胞更多,至缺氧培养12h,大多数细胞破碎。结论:低糖联合缺氧培养模式可以建立可行的O2A细胞外缺氧模型。 Objective: To study the influence of oxygen and glucose deprivation on O2A progenitors. Methods: Trypan blue staining was used to measure O2A progenitors viability. Hochest 33258 fluorescent staining and flow cytometry techniques were used to measure apoptotic cells. Results: The apoptotic O2A cells were not found until the oxygen deprivation combined with low glucose were maintained for 6 h, and promoted in a time-dependent manner. Additional 6 h later, necrosis occured to most O2A cells. Conclusion : Oxygen deprivation combined with low glucose can mimic the ischemic-hypoxic damage of O2A progenitors in vitro.
