摘要
目的研究芎芷地龙汤对偏头痛模型中脑导水管周围灰质c-fos和NOS蛋白共存表达的影响。方法健康雄性SD大鼠随机为4组:生理盐水组、模型组、舒马普坦组、中药治疗组。皮下注射NTG注射剂复制实验性偏头痛动物模型,经药物干预后在4小时、6小时两个时间点取材,采用免疫组化双标染色法测定导水管周围灰质c-fos和NOS的免疫阳性神经元的兴奋状态及其共存表达情况。结果模型组c-fos阳性细胞数和NOS阳性细胞数增多,4小时多于6小时,两个时间点的阳性细胞数与生理盐水组比较有显著性差异(P<0.01)。中药组和舒马普坦组c-fos阳性细胞数和NOS阳性细胞数减少,两个时间点的阳性细胞数与模型组比较有显著性差异(P<0.01)。结论芎芷地龙汤可以减少由硝酸甘油诱导的致痛物质c-fos、NOS的表达,从而进一步减少导水管周围灰质神经元的兴奋性,阻止痛觉信息的传递。
objective To study the effect of Xiongzhidilong-Decoetion on coexistence express of e-fos and NOS in pefiaqueductal gray matter of migraine model. Methods Healthy male SD rats were randomly divided into 4 groups: normal sodium control group, model control group, Sumatriptan control group, Xiongzhidilong-Decoetion group. Reproduce migraine animal model with subcutaneous injection NTG, neuronal emotional state and coexistence express of c-los and NOS which located at periaqueductal gray matter were determined with immunohistoehemical double staining techniques after medicine interfered 4h and 6h. Results Positive neurons of e-fos and NOS of model group were increased, which were significant compared with normal sodium group at 4h and 6h( P 〈 0. 01 ) , the neurons at 4h were more than the contents at 6h. The neurons of c-fos and NOS of Chinese materia medica group and Sumatriptan group were decreased, which were significant compared with model group at 4h and 6h ( P 〈 0. 01 ). Conclusion Xiongzhidilong-Decoction can reduce the expression of c-fos and NOS which were induced by NTG. This effect can depress neuronal excitability in periaqueductal gray matter, and can inhibit nociceptive pain information transmitting.
