摘要
目的通过中药"健身丹"对脊髓型颈椎病神经细胞凋亡相关基因表达影响的研究,从分子生物学角度探讨中医药防治脊髓型颈椎病的机理。方法选用SD大鼠36只,体质量(250±50)g,随机分组。中药治疗组12只,空白对照组12只,正常对照组12只。采用创新大鼠自体第7颈椎棘突椎间移植法制造大鼠脊髓压迫模型,并采用改进Rivlin的斜板实验检验模型是否成功。中药治疗组于造模成功后给予"健身丹"制成粉末悬液灌胃给药2.66g·kg-1.d-1。TUNEL法神经细胞凋亡率检测,Bcl-2mRNA测定。结果相比于空白对照组及正常对照组,中药治疗组大鼠受压节段脊髓抗神经细胞抗凋亡基因Bcl-2mRNA的表达明显提高,组织修复增强,神经细胞凋亡减少。主观症状、体征观察也显示出相似作用。结论中药"健身丹"确实能提高大鼠受压节段脊髓抗神经细胞抗凋亡基因Bcl-2mRNA的表达,促进组织修复,减少神经细胞凋亡。
Objective To explore the mechanism of cervical spondylotie myelopathy at the level of molecular biology and establish the theory foundation of new medicine development by the studying the effects of the traditional Chinese medicine Jianshen Pill on the cervical spondylotic myelopathy at the level of neuroeyte apoptosis Genes. Methods 36 SD rats were divided into 3 groups which were Chinese medicine group(n= 12), blank control group (n= 12), and normal control group (n= 12). The blank control groups and the Chinese medicine group will be established as a cervical spondylotic myelopathy rat model, and the normal control group will be fed as normal. The rat models were established by a bone graft from self 7th cervical vertebrae spinous process at the part of C4-5 or C5-6 intervertebral space. And the rat models were checked whether they were succeeded according to the inclined test ameliorated Rivlin. Results The Chinese medicine group will be drenched with the Jianshen Pill 2.66g/ kg/d. The apoptosis neuron cells were detected by TUNEL and the Bcl-2 mRNA method. Conclusion The Chinese medicine Jianshen Pill could enhance the expression of Bcl-2 mRNA, promote the neuron tissue repair, and incline the neuroeyte apoptosis. Similarly, it could improve the subjective symptoms.
