摘要
从880只250日龄雪山草鸡中选取处于产蛋期、就巢期和恢复期的鸡各20只,屠宰采集下丘脑、垂体、输卵管和卵巢,用荧光实时定量PCR法检测其中催乳素基因mRNA的表达水平,并与繁殖性能进行相关分析,结果表明:PRL基因在4个组织中都有表达,但在垂体中的表达极显著高于其他组织(P<0.01),而且在其他3个组织之间PRL的表达差异并不显著(P>0.05);就巢期鸡PRL基因在垂体中的表达量显著高于产蛋期鸡和恢复期鸡(P<0.05),而在下丘脑、输卵管和卵巢3个组织中3组个体PRL基因的表达量差异不显著(P>0.05);PRL在垂体的表达与种鸡的繁殖性能呈负相关,相关系数为-0.328(P<0.05)。
20 chickens in laying, broodiness and nonovulatory periods, respectively selected from 880 Xueshan chickens aged 250 days, were slaughtered in order to get hypothalamus, pituitary, oviduct and ovarian, in which the mRNA expression level of prolactin gene was detected by real- time PCR. Correlative analysis was carried out between prolactin gene mRNA expression level and the reproductive performance. The results showed that PRL gene was expressed in all of four tissues, but the expression in pituitary were significantly higher than that in other tissues. The expression in the other three tissues was not significantly different. In pituitary, PRL mRNA ex- pression in broodiness period was significantly higher than that in ovulatory and nonovulatory, while in hypothalamus, oviduct and ovarian, the expression didn't show any significant differ- ence. PRL expression in pituitary negatively correlated with reproductive performance of the breeding bird and the correlative coefficient was --0. 328.
