摘要
研究了生长激素(STH)和胰岛素(Insulin)对体外培养猪COCs卵母细胞成熟及孤雌激活后卵裂、卵丘细胞凋亡及凋亡相关基因Bcl-2、Bax表达的影响,结果表明:STH和Insulin对卵母细胞的成熟和激活后卵裂具有双重效应,培养液中添加适当浓度的STH(0.15μg·mL-1)或Insulin(5μg·mL-1)可提高卵母细胞成熟率和卵裂率,与对照组及其他试验组相比差异显著(p<0.05)。在猪COCs体外成熟过程中,适当浓度STH和Insulin能抑制卵丘细胞凋亡,抑制Bax在卵丘细胞的表达。
Effects of somatotropin and Insulin on oocytes of porcine in vitro maturation, cleavage after parthenogenetic activation, apoptosis of cumulus arid expression of Bcl-2 and Bax were studied. The results showed that somatotropin and Insulin had double effect on oocytes maturated in vitro and cleavage. The rate of maturation and cleavage of porcine oocytes matured in medium supplemented with somatotropin (0.15μg·mL^-1) or insulin (5μg·mL^-1) was significantly higher compared with control and other groups(p〈0.05). Somatotropin and Insulin could inhibited apoptosis of cumulus and expression of Bax on cumulus in porcine in vitro maturation cumulus-oocyte complexes.
