摘要
Vsx1基因是与金鱼眼睛空间模式形成相关的调控基因和二倍体依赖型机制的侯选目标基因.为进一步探究Vsx1在金鱼胚胎发育过程中的功能及其调控方式,采用反义寡核苷酸技术抑制Vsx1的表达,应用双向凝胶电泳(2-DE)进行蛋白分离,经PDQuest软件图谱分析,质谱分析初步鉴定,获得了Vsx1沉默引起的13个差异蛋白质,如Prohibitin-2(PHB2)、Nme2 protein等,并对这些差异蛋白质可能直接或间接地受到Vsx1的调控机理和Vsx1在金鱼胚胎发育过程中的功能进行了初步的理论分析,为进一步阐明Vsx1的功能和调控机制奠定了良好的基础.
Vsx1 is a regulatory gene related with spatial patterning of eyes of goldfish and diploid-dependent regulatory mechanism. In order to investigate the function of Vsxl in the development of goldfish embryos and the regulatory pattern, a morpholino antisense oligos inhibition experiment with microinjection was performed to the silence of Vsxl. Proteins were separated with two-dimensional gel electrophoresis (2-DE) and then analyzed with PDQUEST software, Thirteen differential expression protein pots due to the silence of Vsxl were identified by mass spectrometry, such as Prohibitin-2 (PHB2) and Nme2 protein. The possible pattern of the differential expression proteins regulated by Vsxl directly or indirectly and the function of Vsx1 in the development of goldfish embryos were analyzed primarily in theory. The results might be helpful in further investigating the function and the regulatory pattern of Vsx1. 3figs., 2tabs., 17refs.
