原花青素对局灶性脑缺血后凋亡基因Fas表达的影响 被引量:1

摘要 目的探讨原花青素(Procyanidins,PC)对局灶性脑缺血大鼠脑组织中凋亡基因Fas表达的影响。方法随机将30只大鼠均分为假手术组、模型组、PC-1组(50 mg/kg)、PC-2组(100 mg/kg)和PC-3组(200 mg/kg)。各组术前2周及1 h灌胃给予相应药物后以线栓法制备持续性局灶性脑缺血模型,24 h后断头取脑观察脑组织形态,并用免疫组化SABC法染色,统计大鼠海马CA1区每高倍镜视野Fas阳性表达率[阳性细胞数/(阳性细胞数+阴性细胞数)×100%]。结果①脑组织病理变化:PC各剂量组与模型组相比,海马CA1区神经细胞存活数明显增多[(75.3±4.1)/HP、(76.3±5.0)/HP、(82.5±7.1)/HP,(62.8±6.8)/HP,P<0.01]。②免疫组化结果显示PC组可以下调Fas阳性表达率,与模型组比较有显著性差异(P<0.01)。结论PC对局灶缺血性脑损伤具有保护作用,其保护作用机制可能与抑制Fas活性,从而减少细胞凋亡有关。 Objective To study the effects of procyanldlns on expression of Fas in focal cerebral ischemia. Methods Randomly and equally , thirty rats were divided into the sham-operative group, focal cerebral isehemic group and procyanidins groups of 50 mg/ kg, 100 mg/kg and 200 mg/kg. Embolization thread was inserted into all the rats except those in the sham-operative group for the establishment of model of middle cerebral artery occlusion. After the rats in PC-treatment groups were provided with corresponding doses of procyanidins. 2 weeks and 1 hour before MCAO by means of immunohistochemical SABC under high power lens, histopathological changes were observed, as well as the immunohistochemistry changes of Fas. Results (1)Histopathological changes : compared with ischemia group, the number of survival neurons in the CA1 sector of the hippocampus of PC-treatment groups increased evidently [ (75.3 ±4. 1) /HP, (76.3±5.0) / HP, (82.5 ±7. 1) / HP, (62. 8±6. 8) /HP, P〈0.01] . (2)Immunohistochemistry of Fas showed that the positive rates of PC treatment groups were substancially less than that of model group (P〈0. 01). Conclusions Procyanidins plays a neuroprotective role in the focal cerebral ischemia which may be achieved by decreasing the positive expression of Fas.
