AP-2α在小鼠血管内皮细胞中上调VEGF的表达

摘要 转录因子AP-2α(activating protein 2α)在哺乳动物发育、分化以及肿瘤的发生等生命现象中起重要作用,最近的研究发现,在HaCaT角化细胞中,AP-2α可以影响vaseular endothelial growth factor(VEGF)的表达.对C 57BL/6J小鼠胸主动脉血管内皮细胞进行了原代培养,取2~4代细胞分别以转染质粒pCMV-Myc,pCMV-Myc-AP-2α,pCMV-Myc-VEGFA以及SiRNA-AP-2α,Control-RNAi-AP-2α,SiRNA-VEGFA,Control-RNAi-VEGFA.然后采用实时定量RT-PCR和Western Blot分别检测AP-2α和VEGF mRNA与蛋白的表达水平.结果发现在小鼠血管内皮细胞中AP-2α的过表达可使VEGF mRNA和蛋白质表达水平上升;培养细胞转染SiRNA-AP-2α后可使VEGF mRNA的表达和蛋白的表达水平下降;而VEGF的过表达及SiRNA-VEGFA对AP-2αmRNA和蛋白的表达水平没有明显影响,因此,在小鼠血管内皮细胞中AP-2α可上调VEGF的表达. Transcription factor activating protein 2α (AP-2α)regulates transcription of a number of genes involving mammalian development, differentiation and carcinogenesis. Recent studies have showed that AP-2α could affect the expression of vascular endothelial growth factor (VEGF) in HaCaT keratinocytes. The primary cultures of mouse vascular endothelial cells from C57BL/6J mouse were successfully obtained, and then transfected with the plasmid pCMV-Myc, pCMV-Myc-AP-2α, pCMV-Myc- VEGFA, SiRNA-AP-2α, Control-RNAi-AP-2α, SiRNA-VEGFA and Control-RNAi-VEGFA to the primary culture cells, respectively. The level of mRNA and the protein of VEGF were then investigated using quantitative real-time RT-PCR and Western blot, and found that AP-2α could increase the endogenous VEGF expression in mouse vascular, endothelial cells, while the VEGF had not effected on the expression of AP-2α.
