摘要
一种新型的重组溶瘤腺病毒Ad-CEA-ZD55-ST13被成功构建。该病毒利用CEA(carcinoembryonic antigen)启动子和ST13基因特异性增强ZD55系统的肿瘤靶向性。实验表明,重组溶瘤腺病毒Ad-CEA-ZD55-ST13能在结肠癌细胞株SW620中增殖;Ad-CEA-ZD55-ST13增加ST13基因在HCT116细胞中的表达量;Ad-CEA-ZD55-ST13比对照病毒Ad-CEA-ZD55-EGFP对HCT116的杀伤作用强。
In this study, a novel recombinant oncolytic adenovirus Ad-CEA-ZD55-ST13 is successfully constructed, which makes use of the tumor specificity of CEA （carcinoembryonic antigen） promoter and ST13 to enhance the targeting ability of ZD55. It is demonstrated that the recombinant oncolytic adenovirus Ad-CEA-ZD55-ST13 can proliferate in SW620 cells, expression of ST13 gene increases in HCTll6, Ad-CEA-ZD55-ST13 makes more cell killing than Ad-CEA-ZD55-EGFP in HCT116 cells.
