摘要
目的探讨单核细胞趋化蛋白-1(MCP-1)在动脉粥样硬化(AS)大鼠动脉平滑肌细胞(SMC)中的表达及其与AS发生的关系。方法SD雄性大鼠20只随机分为对照组和高脂饮食组,每组10只。对照组用普通饲料喂养,高脂饮食组用高脂饲料喂养,喂养2个月后心脏采血处死大鼠,取胸主动脉,常规固定,包埋,切片。采用免疫组织化学检测各组动脉SMC中MCP-1蛋白的表达情况。结果对照组动脉SMC中MCP-1呈弱阳性表达,高脂饮食组动脉SMC中MCP-1呈强阳性表达。高脂饮食组动脉SMC中MCP-1表达的平均灰度值显著低于对照组(P<0.01)。结论高脂血症可能通过诱导血管壁SMC中MCP-1的表达引起AS的发生。
Objective To explore the mechanism of atherosclerosis (AS) through examining the expression of monocyte chemoattractant protein-1 ( MCP-1 ) in arterial smooth muscle cells (SMC) of rats with atherosclerosis. Methods Twenty male SD rats were randomly divided into control group and high fat diet group, ten rats in each group. The rats of control group were fed with normal diet, and the rats of high fat diet group were fed with high fat diet. Two months later, the rats were executed by being taken out the blood from their heart. The aorta tissue of rats in each group were incised, fixed, embedded and bladed routinely. The expression of MCP-1 protein in smooth muscle cells were detected by immunohistochemistry. Results The expression of MCP-1 in SMC of control group were weaker than that in high diet group. The value of average grey degree of MCP-1 of smooth muscle cells in high fat diet group was obviously lower than that in control group ( P 〈 0. 01 ). Conclusion Hyperlipemia may lead to AS by stimulating the expression of MCP-1 in SMC.
