摘要
用Berthlot法测定酸性脲酶酶反应过程产物NH+4的浓度变化。实验表明,酸性脲酶反应是按Michaelis机理进行。其最佳反应条件是pH=4.5,反应温度60℃,反应活化能Ea=37.34kJ/mol,米氏常数Km=0.2868mmol/L,Vm=0.1744mmol/L·min.
Berthlot method was used to determined the change of ammonia ion concentration during the reaction process.Results show that acid urease catalysed reaction is of Michaelis reaction .It's optimal reaction conditions are as follows:pH=9.0; temperature t=30℃,the energy of activation Ea=37.34 kJ/mol;the Michaelis constant Km=0.286 8 mM,and the maximum velocity is 0.174 4 mM/min.
