右美沙芬对慢性吗啡耐受小鼠脊髓STAT3表达的影响 被引量:1

摘要 目的观察吗啡耐受形成过程中腹腔注射右美沙芬对小鼠腰段脊髓背角STAT3表达的影响,探讨STAT3与吗啡耐受的关系。方法清洁级昆明种小鼠24只,体质量18~22g,随机分为g组,每组6只,A组(对照组):皮下注射生理盐水10mL/kg,30min后腹腔注射生理盐水20mL/kg,每天重复2次,连续9d;B组(慢性吗啡耐受模型组):皮下注射吗啡10mg/kg,30min后腹腔注射生理盐水20mL/kg;C,D两组:吗啡用药均同B组,吗啡注射30min后分别腹腔注射50,100mg/kg右美沙芬。各组小鼠隔天第1次注射后1h观测辐射热痛阈值变化。第9天最后1次注射后取小鼠脊髓L4-6节段,采用免疫组织化学方法测定腰段脊髓背角浅层STAT3免疫反应阳性产物的变化。结果与给药第1天比较,B组给药第5,7,9天时最大镇痛效率(MPE)降低,C组和D组给药第7,9天时MPE降低(P〈0.05)。与A组比较,B组STAT3表达上调,MPE升高(P〈0.01)。与B组比较,C组和D组STA13表达下调,MPE升高(P〈0.05)。结论腹腔注射右美沙芬抑制了慢性吗啡耐受模型小鼠腰段脊髓背角STAT3表达,STAT3信号通路的激活参与了慢性吗啡耐受的维持。
