摘要
目的:探讨缺氧对膀胱逼尿肌细胞缝隙连接蛋白(Cx)43表达的影响。方法:体外培养膀胱逼尿肌细胞,用连二亚硫酸钠清除培养基内的氧,建立缺氧模型,通过RT-PCR、Western blot检测正常对照组、缺氧(A)组及VitE预处理(B)组的逼尿肌Cx43表达情况,并检测丙二醛质量浓度。结果:A、B组Cx43和Cx43mRNA相对表达及丙二醛质量浓度均较对照组明显升高(P<0.01),A组表达最高,与B组比有统计学意义(P<0.05)。结论:缺氧可诱导逼尿肌细胞Cx43高表达,且与氧化损伤程度相关。
Objective: To investigate the influence of hypoxia on connexin(Cx)43 in detrusor smooth muscle cells. Methods: Hypoxia model of detrusor smooth muscle cell in vitro was established by sodium hyposulfite which consumed the oxygen of the medium. The Cx43 and Cx43 mRNA were detected by Western blot and RT-PCR. The level of malon- dialdehyde was detected. Results: The Cx43, Cx43 mRNA and the level of malondialdehyde of group A and B were much higher than those of control group( P 〈 0.01 ), and the expression of group A was higher than that of group B( P 〈 0. 05). Conclusion: Hypoxia can upregulate the expressions of Cx43 and Cx43 mRNA in detrusor smooth muscle cell, and it may be associated with the degree of oxidative damage.
