摘要 目的:探讨增殖细胞核抗原(PCNA)和细胞周期依赖性激酶抑制剂p21 WAF1/CIP1蛋白在大鼠肾上腺细胞体外原代培养中的表达及其意义。方法:用SP免疫组化法检测大鼠肾上腺细胞培养第3、5、7、9、11 d的PCNA和p21蛋白表达强度。结果:在肾上腺细胞体外培养中,第3 d((18±3)%)、5 d((38±5)%)、7 d((70±65)%)PCNA蛋白表达逐渐增强,第9((50±3)%)、11 d((39±2)%)与第7 d比减弱(P<0.05)。第3 d((43±5)%)、5 d((35±2)%)、7 d((21±3)%)p21 WAF1/CIP1蛋白表达逐渐减弱,第9 d((39±3)%)、11 d((52±4)%)与第7 d比重新增强(P<0.05)。结论:肾上腺细胞体外原代培养在第7 d PCNA蛋白表达最强,p21 WAF1/CIP1蛋白表达最弱,肾上腺细胞增殖也最旺盛。 Objective: To explore expression and significance of proliferating cell nuclear antigen(PCNA)and cyclin dependent kinase inhibitor p21WAF1/CIP1 in rots adrenocortical cell culture in vitro. Methods: Immunohistochemical method was adopted to examine the expressions of PCNA and p21WAFI/CIP1 in cell culture at d3, d5, d7, d9 and dll. Results: In adrenoeortical cell culture /n vitro, the expression of PCNA was gradually enhanced at d3((18 ± 3)%), d5((38 ± 5)%)and d7((70 ± 65)%), but were gradually decreased at d9((50 ± 3) %)and d11((39 ± 2)%) ( P 〈 0.05), and the expression of p21WAF1/CIP1 was decreased at d3( (43 ± 5) % ), d5( (35 ± 2) % )and 7d( (21 ± 3)%), but were gradually enhanced at d9((39 ± 3)%)or d11((52±4)%)(P 〈0.05). Conclusion: The proliferation of adrenoeortical cells is the most vigorous in vitro at d7.
