RT-PCR Detecting Expression of Ghrelin in the Antral Follicles of Sheep Ovary

为了明确Ghrelin在绵羊卵巢有腔卵泡上是否存在表达,本研究利用实时定量RT-PCR技术检测了绵羊卵巢有腔卵泡内的卵母细胞、卵丘细胞和壁层颗粒细胞的Ghrelin蛋白的表达量情况。结果揭示绵羊卵巢有腔卵泡内各类型细胞Gh-relin mRNA的相对表达量大致相同。绵羊卵巢有腔卵泡内各类型细胞Ghrelin蛋白的表达及Ghrelin mRNA的表达模式,尤其是卵母细胞中的表达,揭示这一新型分子在绵羊卵巢上具有潜在的调控作用。

The possibility of Ghrelin expressed in antral follicles of sheep ovary was investigated in this study, Ghrelin pro- tein was detected in cumulus cells （CCs） and granulosa cells （GCs）. Relative real-time reverse-transciptase （RT）-PCR experi- rnents confirmed that the Ghrelin mRNA levels did not show significantly difference. The persistent presence of the Ghrelin sig- naling system within antral follicles, including oocytes, CCs and GCs, revealed the possibility of a potential regutatory role of this novel molecule during oocyte maturation and embryonic development.
