摘要
目的:比较茶多酚与艾司洛尔对缺氧联合Ang-2致内皮细胞分泌内皮素(Endothelin,ET)的保护作用。方法:将培养的血管内皮细胞分为4组,分别为①对照组;②缺氧+Ang-2组;③缺氧+Ang-2+茶多酚组;④缺氧+Ang-2+艾司洛尔组,每组8个样本,用放射免疫法测定作用前、作用后0.5、6、24h各组内皮素含量。结果:①缺氧+Ang-2组与对照组比较血管内皮素明显升高(P<0.01);②缺氧+Ang-2+茶多酚组同缺氧+Ang-2组比较血管内皮素显著降低(P<0.01);③缺氧+Ang-2+艾司洛尔组同缺氧+Ang-2组比较血管内皮素显著降低(P<0.01);④缺氧+Ang-2+茶多酚组与缺氧+Ang-2+艾司洛尔组比较血管内皮素浓度降低在0.5h无差异(P>0.05),但在6h和24h差异有统计学意义(P<0.01)。结论:缺氧联合Ang-2可导致血管内皮细胞分泌内皮素功能增强;茶多酚和艾司洛尔对缺氧联合Ang-2对内皮细胞的损害均有保护作用,茶多酚的保护作用强于艾司洛尔。
Objective: To compare the protective effect of Tea polyphenol with that of esmolol on vascular endothelial cells secreting endothelin induced by hypoxia combined Ang-2. Methods: Vascular endothelial cells were divided into 4 groups that included control group, hypoxia+Ang-2 group, hypoxia+Ang-2+tea polyphenol group, hypoxia+Ang-2+esmolol group, and each group comprised eight samples. The changes of endothelin expression level were measured with radioimmunoassay at the time of 0.5 h,6 h,and 24 h. Results: (1)The level of endothelin expression in hypoxia+Ang-2 group was higher significantly than that in the control group (P〈0.01) ; (2)The level of endothelin expression in the bypoxia+Ang-2+tea polyphenol group was lower significantly than that in hypoxia+Ang-2 group (P〈0.01); (3)The level of endothelin expression in the hypoxia+Ang-2+esmolol group was lower significantly than that in hypoxia+Ang-2 group(P〈0.01 ) ; (4)There was not different in level of endothelin expression between the hypoxia+Ang-2+tea polyphenol group and the hypoxia+Ang-2+ esmolol group at the time of 0.5 h after hypoxia combined Ang-2 (P〉0.05),but there was significantly different at the time of 6 h and 24 h after hypoxia combined Ang-2 (P〈0.01).Conclusion:Both hypoxia combined Ang-2 can induce obvious change of endothelin expression level in endothelial cell and this change can be reduced significantly by tea polyphenol and esmolol, the protective effect of tea polyphenol is more obvious compared with that of esmolol.
