摘要
以河南海棠(Malus hupehensis Rehd.)种胚为材料,应用低温和植物生长调节剂6-BA、GA3对离体培养条件下海棠种胚休眠的解除进行了研究。结果表明,低温处理对解除休眠有显著作用,随着低温处理时间的延长,成苗率提高。不进行低温处理的条件下,在MS培养基中附加一定浓度的植物生长调节剂可以解除河南海棠种胚休眠,其中以2.0 mg/L的GA3与0.2 mg/L的6-BA同时使用效果较好,成苗率达94.1%。低温与植物生长调节剂相结合对解除种胚休眠的效果最好。
The experiments of dormancy of in vitro Malus hupehensis Rehd. Embryo broken by low temperature and plant growth regulators (6-BA, GA3) were studied. The results indicated that low temperature had significant effect on breaking dormancy: with the treatment time of lower temperature prolonged, seedling rate was increased. Without low temperature, dormancy of Malus hupehensis Rehd embryos in MS medium could break by certain concentration plant growth regulators. To applied 2.0 mg/L GA3 and 0.2 mg/L 6-BA simultaneously could have better effect, seedling rate reached to 94.1%. The combination of low temperature and plant growth regulators showed the best effect on the breaking dormancy.
