摘要
利用银染S-SAP技术对苹果富士及其14个芽变品种进行研究,结果发现,筛选出的6对引物中,L12/E7能够将所有富士芽变品种同富士区分开来,其余5对引物能够在芽变品种中区分秋富5号、烟富1、盛放富1和长富1号.由于S-SAP多态性主要由基因组内逆转座子的转座引起,因此富士苹果芽变品种的产生可能与逆转座子的插入有关.
'Fuji' apple and its 14 bud mutants were identified with Silver staining S-SAP (Sequence-specific amplification polymorphism). In six primer pairs screened from 44 primer combinations,L12/E7 could discriminate all the bud mutants from maternal 'Fuji' successfully, other 5 primer pairs could discriminate 'Akifu 5''Yanfu 1' 'Shengfangfu 1' and 'Nagafu 1 ' mutants. The polymorphism in S-SAP mainly comes from the transposition of retrotransposon. The results suggested that the bud mutants generated in 'Fuji' appeared to bederived from retrotransposon insertion.
