小茴香对实验性肝纤维化大鼠细胞因子结缔组织生长因子(CTGF)的影响 被引量:5

摘要 目的:观察小茴香对实验性肝纤维化大鼠结缔组织生长因子(CTGF)的影响,分析小茴香抗肝纤维化的治疗作用并探讨其作用机制。方法:70只Wistar健康雄性大鼠随机分为5组,正常对照组、模型组、秋水仙碱治疗组、小茴香低剂量治疗组、小茴香中剂量治疗组、小茴香高剂量治疗组。采用腹腔注射CCl4及饮用乙醇建立肝纤维化模型,并用免疫组化、酶联免疫吸附法(ELISA)检测CTGF在组织、血清中的水平。结果:HE染色显示,经小茴香治疗后肝细胞脂肪变性程度减轻,纤维化程度好转。免疫组化和ELISA结果显示:正常对照组和模型组有统计学差异(P<0.05)。模型组和治疗组比较均有统计学差异(P<0.05)。小茴香高剂量治疗组降低细胞分泌CTGF的作用高于小茴香中、低剂量治疗组和秋水仙碱治疗组(P<0.05)。结论:小茴香低、中、高剂量治疗组均有降低CTGF的分泌、抑制大鼠肝脏炎症、改善肝脏纤维化的作用。 Objective: To investigate the effect on the connective tissue growth factor (CTGF) in CCl4-induced liver fibrosis rat by the foeniculum vulgare mill. Methods: 70 healthy Wistar rats were divided into 5 groups in random, control group, Model group. Colchicines therapeia group (0.02 mg/day), Foeniculum vulgare mill therapeia groups (low, middle, high). The model of liver fibrosis was found in rats on composite factor. CTGF of liver tissue and serum were detected by the method of Immmuno-histochemistry and ELISA respectively. Results: HE stain showed that fatty degeneration and inflammation are light in foeiculum vulgate mill therapeia group (low, middle, high). The immmunohiso-chemistry staining indicated the amount of the cytokines (CTGF) secreted by the cells could be reduced in C, D, E, F, the number of the positive cells decreased . ELISA showed the amount of the cytokines (CTGF) secreted by the cells could be reduced in C, D, E, F. There were significant different between A and B (P〈0.05)and significant different among B and C, D, E, F (P 〈0.05). The high group was better than low and middle group in reducing CTGF. Conclusion: Foeniculum vulgare mill could reduce the CTGF secreted by cells and inhibit the inflammation in rats liver, protect the hepatic cells from damage induced by CCl4 significantly.
