茶叶、蚕豆对人肾小球系膜细胞分泌IL-8影响的实验研究

Tea and Broad Beans Treat the Synthesis and Release of Interleukin-8 in and from Human Mesangial Cells

目的：研究茶叶和蚕豆等对人肾小球系膜细胞（ｈＭＣ）的增殖和合成、分泌ＩＬ－８的影响。方法：应用体外培养技术、３Ｈ－ＴｄＲ掺入法及ＭＴＴ法观察茶叶和蚕豆等对ｈＭＣ增殖的抑制作用。用ＥＬＩＳＡ夹心法测定ＩＬ－１、肿瘤坏死因子（ＴＮＦ）刺激ｈＭＣ的培养上清中ＩＬ－８和ＩＬ－６的含量，并观察茶叶、蚕豆对ｈＭＣ分泌ＩＬ－６和ＩＬ－８的抑制作用。结果和结论：茶叶和蚕豆等对体外培养的ｈＭＣ的增殖有抑制作用，ＩＬ－１、ＴＮＦ可刺激ｈＭＣ分泌ＩＬ－８和ＩＬ－６，而ＩＬ－８对中性粒细胞和Ｔ细胞有强大趋化作用，ＩＬ－６可促进ｈＭＣ增生。茶叶等可抑制ｈＭＣ分泌ＩＬ－８和ＩＬ－６，从而抑制ｈＭＣ增生。

Objective:To assess the effect of tea and broad beans on proliferation and synthesizer by culture of human mesangial cells(hMC) and of releasing IL-8 and IL-6.Methods:In this study we reported the effect of tea and broad beans agents on hMC proliferation by culture of hMC,3H-TdR incorporation and MTT assay.We have examined that both IL-1 and TNF stimulate hMC to release IL-6 and IL-8 by ELISA.Meanwhile,our results showed that tea and broad beans agents inhibit hMc to release IL-8 and IL-6.Results and conclusion:Our studies identified that these agents could inhibit hMC growth and the generation of IL-8 and IL-6.Thus the results of these studies suggest that these tea and broad beans agents have therapeutic potential for the treatment of glomerulonephritis.
