摘要
目的:研究茶叶和蚕豆等对人肾小球系膜细胞(hMC)的增殖和合成、分泌IL-8的影响。方法:应用体外培养技术、3H-TdR掺入法及MTT法观察茶叶和蚕豆等对hMC增殖的抑制作用。用ELISA夹心法测定IL-1、肿瘤坏死因子(TNF)刺激hMC的培养上清中IL-8和IL-6的含量,并观察茶叶、蚕豆对hMC分泌IL-6和IL-8的抑制作用。结果和结论:茶叶和蚕豆等对体外培养的hMC的增殖有抑制作用,IL-1、TNF可刺激hMC分泌IL-8和IL-6,而IL-8对中性粒细胞和T细胞有强大趋化作用,IL-6可促进hMC增生。茶叶等可抑制hMC分泌IL-8和IL-6,从而抑制hMC增生。
Objective:To assess the effect of tea and broad beans on proliferation and synthesizer by culture of human mesangial cells(hMC) and of releasing IL-8 and IL-6.Methods:In this study we reported the effect of tea and broad beans agents on hMC proliferation by culture of hMC,3H-TdR incorporation and MTT assay.We have examined that both IL-1 and TNF stimulate hMC to release IL-6 and IL-8 by ELISA.Meanwhile,our results showed that tea and broad beans agents inhibit hMc to release IL-8 and IL-6.Results and conclusion:Our studies identified that these agents could inhibit hMC growth and the generation of IL-8 and IL-6.Thus the results of these studies suggest that these tea and broad beans agents have therapeutic potential for the treatment of glomerulonephritis.
