摘要
对簸箕柳组织培养技术进行了初步探索.结果表明:以簸箕柳嫩枝条为外植体,消毒以70%(体积分数)乙醇30-40s+0.1%(质量分数)HgCl2 4-5min为宜;用White为基本培养基,激素组合以6-BA1.0mg·L^-1+NAA 0.01mg·L^-1。诱导腋芽效果最好;BA对腋芽的诱导率有显著影响,回归方程为Y=29.806+126.517X-64.522X^2;基本培养基对丛生芽的诱导无明显影响;不加激素的White和B5培养基均可诱导无菌苗生根.
Tissue culture technique of Salix suchowensis was studied in this paper. The result indicated that the optimum sterilizing conditions were-0.1% mercuric chloride for 4-5 minutes and 70% alcohol for 30-40 seconds by using tender stem with lateral bud as explants. The best phytohormone combination for inducing axilla bud was 1.0 mg·L^-1 of 6-BA+0.01mg·L^-1 of NAA when the basic medium B5 was used. The concentration of BA had remarkable effect on axilla buds induction,and the regression equation is Y=29.806+126.517X-64.522X^2. But the induction of shoot clusters could not distinctly be effected by basic medium. Rooting plants could be induced in whether White or B5 medium with no phytohormone.
