川芎嗪和异博定联合逆转HL60/HT细胞的多药耐药 被引量:15

摘要 观察中药钙通道阻滞剂川芎嗪(TMP)和异博定(VP)对白血病HL60/HT耐药细胞系多药耐药(MDR)的逆转.方法:采用四唑氮蓝(MTT)法作为药物敏感实验,流式细胞仪测量细胞内柔红霉素(DNR)浓度.结果:采用非细胞毒剂量的100mg/LTMP和(或)0.32mg/L及1mg/L的VP与DNR联合应用,均能提高DNR的细胞杀伤作用,其效果比较为VP(1mg/L)+TMP>VP(1mg/L),VP(0.32mg/L)+TMP>TMP>VP(0.32mg/L)(P<0.01),而且VP(1mg/L)的逆转效果与VP(0.32mg/L)+TMP相当.同时用流式细胞仪测量细胞内DNR浓度发现TMP和(或)VP均能增加HL60/HT细胞内DNR的浓度,效果比较VP(1mg/L)+TMP>VP(1mg/L)>TMP.结论:TMP与VP有协同逆转作用,是一种有效的中药白血病MDR逆转剂. Aim:To investigate the reversal of multidrug resistance (MDR) of leukemia cell lines HL60/HT by traditional calcium channel blocker tetramethylpyrazine (TMP)and verapamil (VP). Methods:The drug sensitivity test wasperformed with MTT and the concentration of daunorubicin(DNR) in cells was measured by flow cytometer (FCM).Results: The cytotoxicity of DNR to HL60/HT was enhanced by TMP and (or) VP at nontoxic doses, the effectwere VP (1 mg/L) +TMP>VP (1 mg/L), VP (0. 32 mg/L ) +TMP>TMPM 0. 32 mg/L VP, and the effect of VP(0. 32 mg/L) +TMP was similar to that of VP (1 mg/L).Meanwhile TMP and (or ) VP increased the concentration of DNR in cells. And the effects were VP(1 mg/L) +TMP>VP (1 mg/L) >TMP. Conclusion: For the reversaleffect of VP was enhanced by being combined with TMP,TMP may be a safe and effective revertant to MDR.
