候选抑癌基因Syk与宫颈癌发生发展的相关性研究 被引量:1

Expression of Syk gene and its significance in cervical carcinoma

摘要 目的:探讨脾酪氨酸激酶(spleen tyrosinekinase,Syk)在宫颈癌组织的表达及其临床意义。方法:采用RT—PCR检测60例宫颈癌、50例宫颈上皮内瘤变(CIN)和20例正常宫颈组织中SykmRNA的表达。结果:20例正常宫颈组织和18例CINI组织均检测到Syk基因的表达,32例CINⅡ~Ⅲ组织有17例检测到Syk基因的表达,60例宫颈癌组织有21例检测到Syk基因的表达,Syk基因的表达从宫颈癌组织、宫颈上皮内瘤变组织到正常宫颈组织依次增高(P:0.00),且在CINI中的表达明显高于CINⅡ~Ⅲ,差异有统计学意义,P=0.00。有淋巴结转移的23例宫颈癌组织,有4例检测到Syk基因表达;无淋巴结转移的37例中,17例有Syk基因表达,Syk基因在有淋巴结转移的组织中表达率明显低于无淋巴结转移纽,P=0.02。Syk基因在宫颈癌中的表达与肿瘤淋巴结转移的有明显相关性,但与患者年龄、肿瘤大小、临床分期、组织学类型及病理类型无明显相关性。结论:Syk基因低表达或表达缺失可能与子宫颈癌发生发展有关,提示可能是宫颈癌的一种候选抑癌基因。 OBJECTIVE: To explore the expression and its clinical significance of Syk (spleen tyrosine kinase) gene in cervi- cal carcinoma. METHODS: By using RT-PCR technique, speci- mens from 60 cervical cancer patients, 50 cervical intraepithelial neoplasia (CIN) patients, and 20 normal cervical tissues were de- tected for their expression of the Syk gene. RESULTS: The ex- pression of Syk gene was detected in 20 normal cervical tissues and 18 CINI samples. Seventeen out of 32 CINⅡ-Ⅲ samples were found the expression of Syk mRNA, and 21 out of 60 cervical carcinoma samples were found the expression of Syk mRNA. The Syk expression in cervical carcinoma was lower than that in nor- mal cervical tissues and CIN samples (P:0.00). The expression of Syk was higher in CINI than that in CINⅡ-Ⅲ (P=0.00). In 23 cases with lymph node metastasis, 4 were found the expres- sion of Syk mRNA; in 37 cases without lymph node metastasis, 17 were found the expression of Syk mRNA, and a significant difference was noted between the two groups, P= 0. 02, while there were no significant correlations with the age, tumor's size, clinical stage, histological size and the pathological types of the tumor. CONCLUSION: The reduced or lost expression of Syk may play an important role in suppressing growth and metastasis of cervical carcinoma, and indicate that Syk may be a candidate tumor suppressor of this kind of carcinoma.
