摘要
探讨促性腺激素释放激素(GnRH)及其受体表达与肝癌病理学分级和肝癌细胞增殖的关系.方法:用常规组织学HE染色确定肝癌的病理学分级;用免疫组织化学SABC技术检测49例肝细胞性肝癌(HCC)中增殖细胞核抗原(PCNA),GnRH及其受体的表达.结果:GnRH及其受体均定位于肝癌细胞的胞膜和胞浆.Ⅰ级~Ⅱ级肝癌组织GnRH的阳性率为84.6%(20/23),显著高于Ⅲ级肝癌47.1%(8/17)的阳性率(P<0.05)和Ⅳ级肝癌11.1%(1/9)的阳性率(P<0.01).Ⅰ级~Ⅱ级肝癌组织GnRH受体的阳性率为91.1%(21/23),明显高于Ⅲ级肝癌41.2%(7/17)的阳性率(P<0.01)和Ⅳ级肝癌11.1%(1/9)的阳性率(P<0.01).GnRH阳性肝癌的PCNA标记指数(20.4±12.1)%低于GnRH阴性肝癌(39.1±25.3)%(P<0.05);GnRH受体阳性肝癌的PCNA标记指数(19.2±13.5)%亦明显低于GnRH受体阴性肝癌(40.5±29.1)%(P<0.01).结论:GnRH可能参与肝癌细胞的增殖和分化的调节.
Aim:To explore the possible relationship of gonadotropin releasing hormone (GnRH) and its receptor(GnRHR) expression with hepatocarcinoma cell proliferation and differentiation. Methods: Proliferating cell nuclearantigen (PCNA),GnRH and its receptor were detected in 49hepatocellular carcinoma (HCC) by immunohistochemicalSABC technique. The pathological degree of HCC wasdemonstrated by hematoxilin and eosin (HE) staining. Results:GnRH and its receptor had the same distribution pattern in hepatocarcinoma cells. The positive signal was foundmainly on the surface and in the cytoplasm of hepatomacells. Edmondson's grade Ⅰ~Ⅱ HCC had a higher GnRHpositive rate (84. 6%; 20/23) than those of grade Ⅲ HCC(47. 1 %; 8/17) and grade Ⅳ HCC (11. 1 %;1/9),the differences were significant (P<0. 05 and P<0. 01 respectively).In grades Ⅰ~Ⅱ HCC GnRHR positive rate was 91. 1 % (21/23),which was higher than 41. 2% (7/17) in grade Ⅲ HCC(P<0.01) and 11.1 % (1/9) in grade Ⅳ HCC (P<0. 01 ).The PCNA labeling index was (20. 4±12. 1 ) % in GnRHpositive HCC and (39.1±25. 3) % in GnRH negative HCC,the difference was significant (P< 0.05 ); similarly, GnRHRpositive HCC also had a lower PCNA labeling index(19.2±13. 5 ) % than that of GnRHR negative HCC (40. 5±29. 1 ) %(P<0. 01 ). Conclusion: GnRH might be involved in theregulation of human hepatocarcinoma cell proliferation anddifferentiation.
