胰腺导管癌细胞表达嗜铬颗粒素C/分泌颗粒素Ⅱ和胰抑素 被引量:5

Expression of chromogranin C/secretogranin Ⅱ and pancreastatin in pancreatic ductal carcinoma

摘要 目的确定嗜铬颗粒素C/分泌颗粒素Ⅱ(CgC/SgⅡ)和胰抑素(PS)在胰腺导管癌细胞中的表达.方法采用荧光双标记免疫组织化学技术结合激光扫描共聚焦显微镜观察胰腺导管癌(1996/1997,n=10)细胞中CgC/SgⅡ和PS的表达.结果在CgC/SgⅡ单克隆抗体染色后发现,胰腺导管癌的大多数癌细胞呈CgC/SgⅡ免疫反应阳性,免疫反应阳性物质位于细胞胞质内,而细胞核为阴性.CgC/SgⅡ免疫阳性物质在胞质内为均质性或颗粒状,在一些腺管的癌细胞内存在较大的颗粒状荧光物质.同时用PS抗体的染色发现,在高分化胰腺导管癌CgC/SgⅡ免疫阳性的细胞大部分同时表达PS,但在中分化胰腺导管癌CgC/SgⅡ免疫阳性的细胞仅有一部分细胞表达PS.而且PS阳性物质在高分化胰腺导管癌细胞多为均质性,而在中分化胰腺导管癌细胞为均质性和颗粒状荧光,一些细胞的颗粒状荧光物质仅呈CgC/SgⅡ免疫阳性.结论胰腺导管癌细胞中存在嗜铬颗粒素C和胰抑素. AIM To determine the expression of chromogranin C/secretogranin Ⅱ(CgC/SgⅡ) and pancreastatin (PS) in the pancreatic ductal carcinoma. METHODS Immunohistochemical method combined with laser scanning confocal microscopy was used. RESULTS Monoclonal antibody against CgC/SgⅡ stained most, and possibly all, of the tumor cells in tumor tissue. The intense fluorescence was present in cytoplasm and negative in the nucleus. The immunoreactive CgC/SgⅡwas homogeneous and granular in the cytoplasm. The large granular substances were observed in some tumor cells. Immunostainning with polyclonal antibody anti PS antibody simultaneously was detectable in almost all of the CgC/SgⅡ positive neoplastic tissue sections of the well differentiated pancreatic ductal adenocarcinoma, but only small amount of CgC/SgⅡ positive cancer cells were PS positive in the moderately differentiated pancreatic ductal adenocarcinoma. The immunoreactive PS was homogeneous in the tumor cells of well differentiated pancreatic ductal adenocarcinoma and granular or homogenous in the tumor cells of the moderately differentiated pancreatic ductal adenocarcinoma. The granular fluorescent substances of some tumor cells was immunostained for CgC/SgⅡ only. CONCLUSION CgC/SgⅡ and PS can express in the tumor cells of the pancreatic ductal adenocarcinoma.
