清肺口服液对3I、7b型腺病毒感染人胚肺成纤维细胞TGF-β_1、PDGF-BB mRNA基因表达的影响 被引量:1

摘要 目的观察清肺口服液含药血清对腺病毒3I、7b感染的人胚肺成纤维细胞转化生长因子-β1(TGF-β1)、血小板衍生生长因子-BB(PDGF-BB)mRNA基因表达的影响。方法以3I、7b型腺病毒分别攻击体外培养的人胚肺成纤维细胞,制备清肺口服液含药血清,作用于该细胞,另设正常细胞组、病毒对照组、利巴韦林组,采用原位杂交法检测TGF-β1、PDGF-BBmRNA基因的表达,进行各组间比较。结果病毒对照组较正常细胞组的TGF-β1、PDGF-BBmRNA表达均明显升高(P<0.01),含药血清组对3I、7b型攻击后的TGF-β1、PDGF-BBmRNA表达均有显著降低作用(P<0.01)。结论清肺口服液可下调TGF-β1、PDGF-BBmRNA表达,这可能是其抗病毒作用的机制之一。 Objective To study the effect of Qingfei oral liquid contained serum on the transforming growth factor-β1 (TGF-β1) and platelet-derived growth factor-BB (PDGF-BB) mRNA gene expression of human embryonic lung fibroblast cells infected by adenovirus (ADV) type 3I and 7b. Method TGF-β 1 and PDGF-BB mRNA expression of human embryonic lung fibroblast ceUs infected by ADV type 3I and 7b were determined by in situ hybridization before and after treated with Qingfei oral liquid contained serum. Results ADV could up-regulate TGF- β 1 and PDGF-BB mRNA of human embryonic lung fibroblast cells (P 〈 0.01), while Qingfei oral liquid could down-regulate them (P〈0.01, P〈0.05). Conclusions Qingfei oral liquid played an antiviral role by decreasing the TGF-β1 and PDGF-BB mRNA expression.
