摘要
目的:探讨Survivin基因在肝癌细胞中的表达及与bcl-2/bax的关系。方法:体外培养的人肝癌细胞SMMC-7721,Hep-G2及正常肝细胞,采用免疫组化,RT-PCR法及Westernblot法检测Survivin及bcl-2/bax基因的表达。结果:免疫组化法显示肝癌细胞中有Survivin基因表达,且在SMMC-7721比在Hep-G2表达更明显,而在正常肝细胞中Survivin基因表达不明显;Survivin高表达的细胞,bcl-2也增高。Westernblot和RT-PCR法均显示Survivin基因的表达在肝癌细胞中增加。结论:Sur-vivin基因与人肝癌细胞SMMC-7721及Hep-G2有关,与肝癌细胞的分型相关,由此推断,Survivin基因与肝癌的发生、发展及预后有关。
Objective: This study was aimed at investigating surviving and bel-2/bax expression in human hepatoma cell SMMC-7721 and Hep-G2 line as well as theirs relative. Methods:Immuno-histologieal assay were applied to detect the expression of surviving and bcl-2/bax genes in the hepatoma SMMC-7721 and Hep-G2 cells. Survivin and bcl-2/bax protein and mRNA expressions were measured with Western blot analysis and reverse transcription polymerase chain reaction (RT-PCR), respectively. Results: Both the Western blot and RT-PCR analyses showed survivin and bcl-2/bax expression in SMMC-7721 cell increasing and in the Hep-G2 cell posteriorly but no expression in the natural liver. Immuno-histological assay revealed the same results with the Western blot and RT-PCR. Conclusions: Survivin gene is related with human hepatoma cell SMMC-7721 and Hep-G2. Suvivin gene may play a role in inhibiting apoptosis in liver cancer and may function importantly in tumor production and development.
