摘要
为确定大孔树脂分离杜仲总黄酮的最佳工艺条件,以吸附率及解吸率为考察指标,确定最佳型号树脂、静态和动态吸附及解吸的相关影响因素。结果表明,D-100型大孔树脂对杜仲总黄酮有良好的吸附性,其吸附分离的工艺条件的药液浓度为0.4 mg.mL-1,以2 mL.min-1吸附速率进行吸附,5倍柱体积70%乙醇,洗脱速率为1 mL.min-1时洗脱效果最佳。该方法简单易行,分离效果好,适于杜仲总黄酮的分离纯化。
To establish the optimum process conditions for purification of flavoniods from Eucommia ulmoides by macroporous resin, Seven types of macroporous resin were used to purify flavoniods of Eucommia ulmoides,which determined the optimum macroporous resin,relative factors of adsorption and desorption for static state and dynamic state through adsorption and desorption rate. The results showed that the D-100 maeroporous resin had good absorbing effect on flavoniods of Eucommia ulmoicles. The absorbing technological environment as follows:the liquid concentration was 0.4 mg·mL^-1, the absorbing speed was 2 mL·min^-1. The elution effect of 70% alcohol of 5 bed volume was the best at the eluting velocity of 1 mL·min^-1. This method was simple and feasible, fit for separating and purifying flavoniods from Eucommia ulmoides.
