摘要
目的建立人血清中7种游离脂肪酸的测定方法。方法人血清中的硬脂酸、油酸、亚油酸、十八碳三烯酸、二十碳四烯酸、二十碳五烯酸和二十二碳六烯酸通过加入正十七酸内标,经硫酸/甲醇甲酯化,正己烷萃取浓缩,气相色谱法定性定量。结果7种脂肪酸甲酯化后可完全分离,重复性好,在5.0~500μg/ml范围内相关系数r>0.999,检出限≤1.1μg/ml,20、100、300μg/ml三个浓度的加标试验平均回收率在91.7%~98.1%之间,RSD1.9%~4.1%之间。结论本法适用于人血清中游离脂肪酸测定。
Objective To establish a method for the determination of 7 free fatty acids in human serum. Methods By adding n heptadecanoic acid as internal standard, stearic acid, oleic acid, linoleic acid, Linolenic acid, arachidonic acid, eicosapentaenoic acid and docosahexaenoic acid in human serum were methylesterified by H2 SO4/methanol, extracted and concentrated by n - hexane, detected by gas chromatography. Results After methyl esterification, 7 kinds of fatty methyl ester could be separated completely, with a good reproducibility. The linearity was ranged from 5.0μg/ml- 500μg/ml (r 〉 0. 999), minimum limited determination was ≤1. 1μg/ml. The average recovery rate of 3 levels (20μg/ml, 100μg/ml, 300μg/ ml) was between 91.7 % and 98.1%, RSD was from 1.9 % to 4.1%. Conclusions The method can be used to the determination of free fatty acids in human serum.
