磷酸喹哌iv9mg/kg明显降低氯化钙所致小鼠室颤的死亡率;ip18mk/kg对氯仿诱发小白鼠室颤具有保护作用;iv3.4mg/kg明显缩短肾上腺素诱发家兔心律失常的持续时间;iv6.3mg/kg显著增加恒速滴注乌头碱引起麻醉大鼠室性早搏(VE)、定性心动过速(VT)、室性纤颤(VF)所需的乌头碱用量;iv5.4mg/kg显著增加恒速滴注哇巴因引起麻醉豚鼠VE、VT和VF所需的哇巴N用量;iv3.4mg/kg对麻醉家兔心脏功能和血流动力学的影响较小:CO、MAP呈一过性减少3min内恢复正常,士dp/dtmax、LVE-DP、CI、SI、LVMI均未见明显改变,TPVR也无明显降低。
Piperaquine Phosphate(PQP) iv 9mg/kg markedly decreased the mortality of mice with ventricular fibrillation(VF)induced by CaCl2,PQP,ip 18mg/kg prevented mice from chloroform induced VF in the anesthetized rats,PQP iv 6.3mg/kgsignificantly elevated the dose of aconitine required to induce ventricular extrasystole (VE),ventricular tachycardia (VT) and VFPQP iv 5.4mg/kg dramatically increased the dose of ouabain required to induce VE,VT and VF in the anesthetized guinea pigs andPQP iv 3.4mg/kg greatly shortened the duration of arrhythmia elicited by iv epinephrine in unanesthetized rabbits.The resultssuggested that PQP exerted antiarrhythmic action.The iv LD of PQP in mice was 93mg/kg.The effects of PQP on cardiac functionand hemodynamics were studied on anesthetized rabbits.The results showed that this drug iv 3.4mg/kg could reduce thecontractility of cardiac muscles.CO and BP showed a transient decrease and recovered in 3 minutes.In a few words,PQP has acertain antiarrhythmic effect,but only has a little effect on cardiac function with low toxicity.PQP is found to be a prospectiveantiarrhythmic drug.
