川芎嗪对百草枯中毒大鼠急性肺损伤的治疗作用 被引量:5

摘要 目的探讨川芎嗪对急性百草枯(PQ)中毒大鼠所致肺损伤的治疗作用。方法采用腹腔注射20%PQ溶液制作大鼠急性PQ中毒模型,将大鼠随机分成空白组、阴性对照组、阳性对照组、川芎嗪低剂量组和川芎嗪高剂量组,观察大体标本、组织病理以及肺湿/干质量比、肺泡灌洗液(BALF)中性粒细胞比和蛋白含量。同时测定血清和BALF中MDA含量、SOD和GSH-Px活力以及肺组织核因子-кB(NFк-B)和iNOS活性。结果与阴性对照组相比,川芎嗪低剂量组肺组织淤血、水肿明显减轻,血浆和BALF中MDA含量明显下降(P均<0.01);GSH-Px和SOD活力明显升高(P<0.01)。NF-кB和iNOS降低(P<0.05,或<0.01)。结论川芎嗪可使急性PQ中毒引起的脂质过氧化损害得到改善,且能降低NFк-B及iNOS水平,减轻PQ中毒大鼠肺组织损伤。 Objective To study the therapeutic effect of ligustrazine against acute lung injury by paraquat (PQ). Methods 50 SD rats were randomly divided into five experimental groups:blank group, negative control group, positive control group, ligustrazine low-dose group(LDG) and figustrazine high-dose group(HDG). Macroscopic and histopathological examinations were performed and biological indexes were measured for the lung specimens. The indexes included lung wet weight/dry weight, the rats of nentrophils and protein content in the pulmonary alveolar lavage fluid. In the mean time, the levels of MDA, superoxide dismutase(SOD), glutathione peroxidase(GSH-Px) both in plasma and bronchoalveolar lavage fluid(BALF) were detected. The activities of NK-κB and iNOS in lung tissue homogenate were measured. Results Compared with negative control group, lung congestion and lung edema of ligustrazine low-dose group were mitigated. The level of MDA decreased in both plasma and BALF( P 〈0.01 ), and the SOD and GSH-Px activities were increased ( P 〈 0.01 ). The activities of NK-κB and iNOS in lung tissue homogenate were significantly decreased ( P 〈0.05, 〈0.01 ). Conclusions Administration of ligustrazine could improve the lipid peroxidation damage and ameliorate the lung injury and decrease the concentrations of NK-KB, iNOS caused by acute poisoning of PQ.
