红花黄色素对实验性肾间质纤维化大鼠TGF-β及Ⅰ型胶原表达的影响 被引量:11

摘要 目的观察红花黄色素注射液对肾间质纤维化大鼠TGF-β及Ⅰ型胶原表达的影响,并探讨其肾脏保护机制。方法45只雄性SD大鼠,随机分为假手术组(A组)、单侧输尿管梗阻(Unilateral ureteral obstruction;UUO)组(B组)、UUO+红花黄色素治疗组(C组),各15只。C组红花黄色素5 mg/kg.d,造模术前1天开始腹腔注射给药,各组术后10 d处死大鼠,处死后取术侧肾组织行HE、Masson染色,转化生长因子(Transforming growth factor-β;TGF-β1),Ⅰ型胶原(CollagenⅠ,ColⅠ)免疫组化检测、结果应用图像分析系统进行半定量分析。结果B组、C组可见明显肾间质纤维化的病理改变。C组与B组相比,肾间质纤维化病变程度较轻。免疫组化结果提示:C组与B组相比TGF-β1、ColⅠ表达减少(P<0.01)。结论注射用红花黄色素对大鼠肾间质纤维化有保护作用,这种作用可能是通过抑制TGF-β1的表达,减少Ⅰ型胶原合成的作用实现的。 Objective To observe the effects and mechanism of Safflow Yellow (SY) on TGF-β and Col Ⅰ in rats with renal interstitial fibrosis. Methods 45 male sprague-dawley rats were randomly divided into 3 groups: sham-operated group (A), the UUO model group (B) and the treatment group treated with Safflow Yellow injection (C). Safflow Yellow injection 5mg/kg·d was administered to rats day before the surgery. Kidneys were collected after 10 days. Histological changes through abdominal cavity 1 were also observed with HE and Masson stain. TGF-β,Col Ⅰ were examined with immunohistochemical staining. Results Marked renal interstitial fibrosis changes were found in group B and group C. The lesion area in kidneys of group C significantly decreased compared with that of group B (P〈0.01). By using immunohistological staining, the protein level of TGF-β1 and Col Ⅰ in B group was various, in group C the protein level of TGF-β1 and Col Ⅰ was significantly lower than that of group B (P〈0.01). Conclusion Safflow Yellow could ameliorate renal interstitial fibrosis in rats,which might be realized by reducing the expression of transforming growth factor-β1 and reducing collagen Ⅰ protein synthesis.
