金复康口服液对耐药人肺腺癌A549/DDP膜转运蛋白LRP MRPmRNA表达的影响

Effect of Jin Fu Kang Oral Liquid on the Expression of MRP mRNA-LRP mRNA of the Human Lung Adenocarcinoma A549/DDP Line Cell

目的:从逆转多药耐药的角度,研究金复康口服液对耐药人肺腺癌细胞A549/DDP的化疗增效的作用和分子机制。方法:应用血清药理学方法,选择A549、A549/DDP细胞,实验分为实验组和空白对照组,从细胞水平、分子水平进行研究。结果:金复康口服液低剂量能够逆转A549/DDP对DDP的耐药,逆转耐药倍数为3.45倍,与DDP(320μmol/L)合用,使DDP(320μmol/L)对A549/DDP细胞的抑制率由47.55%升高为73.32%,且q>1.15,表明金复康口服液与DDP合用有明显的协同增效作用,增加了DDP对A549/DDP细胞的敏感度;金复康口服液低剂量能够降低A549/DDP膜转运蛋白LRP、MRP的mRNA表达。结论:(1)低剂量金复康口服液与DDP合用能够增加DDP对A549/DDP的增殖抑制作用,二者具有显著的协同增效作用。(2)金复康口服液能够降低A549/DDP膜转运蛋白LRP、MRP的mRNA表达。金复康口服液可能通过降低耐药肿瘤细胞膜转运蛋白的表达而逆转肿瘤多药耐药,为扶正方药对化疗增效机理提供了新的实验依据。

Objective： From the context of reversal of muhidrug resistance, the role and molecular mechanisms of Jin Fu Kang Oral Liquid and chemotherapy synergies and efficiency to the human lung adenocarcinoma A549/DDP line cells was studied.Methods：The human lung adenoearcinoma A549/DDP line cells were divided into experimental group and control group at random. Serum pharmacology was applied in cell and molecular level.Result：It showed that Jin Fu Kang Oral liquid low dose drug serum can reverse multidrug-resistance of A549/DDP line cell to DDP,reversing-resistance multiples is 3.45. The combination of Jin Fu Kang Oral liquid low dose drug serum and DDP（320 μmol/L） raise the rate of inhibition of DDP to A549/DDP from 47.75% to 73.32%,and q 〉 1.15, and it shows significant synergies and efficiency. Jin Fu Kang Oral liquid low dose can reduce the expression of MRP mRNA-LRP mRNA of human lung adenocarcinoma A549/DDP line cell.Conclusion：（1） Jin Fu Kang Oral liquid low dose combined with DDP can inhance the inhibition of DDP to A549/DDP line cell., and it shows significant synergies and efficiency.（2） Jin Fu Kang Oral liquid can reduce the expression of MRP mRNA-LRP mRNA of the human lung adenocarcinoma A549/DDP line cell. Jin Fu Kang Oral liquid can reverse multidrug resistance of tumor line cell maybe by reducing the expression of the member transport-protein.These will provide the new basis of TCTM of strengthening the body risistance to anticancer and chemotherapy synergies and efficiency.
