摘要
[目的]将内蒙古传统相马术和现代分子生物学技术相结合,找出一种更为科学简便的相马方法。[方法]对试验马COMT基因位点单个基因的多态性与性格性状关联性进行研究,并从分子生物学角度探讨马的性格遗传特点。[结果]电泳检测表明:COMT基因第2外显子的PCR产物与Marker DL2000相比,其位点在250 bp下方。PCR-SSCP分析结果表明:马基因组DNACOMT基因第2外显子存在单核苷酸多态性,所检测的30个个体中,发现2种基因型,即AA和AB型。测序结果表明:COMT基因的第2外显子161位置存在一个多态位点。COMT基因第2外显子多态性与马的竞争性有显著相关性,与其它性格没有显著相关性,AB型马的竞争性比AA型强。[结论]该研究为马的鉴定提供了新的技术支持。
[ Objective ] The research aimed to combine Mongolian traditional sauma method with modem molecuar biotechnology so as to find a more scientific, simple and convenient sanma method. [ Method] The correlation between polymorphism of single genes on COMT gene site of tested horses and their characters and characteristics was researched and the properties of character inheritance in horse were discussed in view of molecular biology. [ Result ] The electrophoresis detection showed that compared with Marker DL2000, the site of PCR products from the 2nd exon of COMT gene was below 250 bp. The results of PCR-SSCP analysis showed that the 2nd exon of COMT gene in horse genome DNA had single nucleotide polymorphism and there were 2 genotypes found in the 30 detected individuals, they were genotypes AA and AB. The sequencing results showed that there was a polymorphic site at the position 161 of the 2nd exon of COMT gene. The polymorphism of the 2rid exon of COMT gene had significant correlation with the competitiveness of horse and had no significant correlation with its other characters. The horse with genotype AB was more competitive than that with AA. [ Conclusion] The research provided new technological support for the identification of horse.
