小鼠骨髓间充质干细胞的体外培养和鉴定 被引量:2

摘要 目的采用全骨髓贴壁培养法分离和体外培养小鼠骨髓间充质干细胞,并通过形态学观察和排除鉴定法进行初步鉴定。方法无菌分离小鼠胫骨和股骨,剪开骨干骺端,用RPMI 1640培养基反复液冲洗骨髓腔,收集并过滤离心。DMEM-LG培养基重悬细胞,将细胞悬液转入0.1%明胶包被的培养瓶中,37℃,5%CO2常规进行原代培养并传代。在培养过程中,取不同代数的细胞做CD45流式标记鉴定。结果采用全骨髓贴壁培养法获得的骨髓间充质干细胞贴壁时间短,增殖快,经传代后能够纯化。第4-5代骨髓间充质干细胞形态单一均匀,呈典型的极性漩涡状生长,并不表达白细胞标志抗原CD45。结论本实验可稳定获得均质性良好的小鼠骨髓间充质干细胞,并初步通过鉴定。 Objective To culture mouse bone marrow mesenchymal stem cells (BMSCs) by whole bone marrow adherence method and assess by morphology and negative surface marker. Methods Mouse limbs were sterilely isolated. Bone marrow was obtained by rinsing with RPMI 1640. After marrow cells harvesting and purifying, they were cultured in vitro. Expression of BMSC negative surface antigens CD45 was detected by flow cytometer in different passages. Results BMSCs isolated by the adherence separation had short adherence time and fast multiplication rate, which could be purified by subculture. At the 4-5th passage, BMSCs showed the typical polar swirl morphology and were negative for CD45. Conclusion BMSCs by the adherence separation method have a good homogeneity. The cells were identified as mouse BMSCs by morphology and negative surface antigens.
