摘要
研究了纤维素酶辅助提取银杏叶总黄酮工艺条件。对银杏叶粒径、纤维素酶用量、酶解时间进行单因素考察,根据单因素考察结果设计正交试验,以总黄酮得率为指标确定适宜的正交试验参数:银杏叶细粉100目、纤维素酶用量30 FPIU/g银杏叶、酶解时间50 min,该工艺条件下银杏叶总黄酮提取得率为2.51%,与未添加纤维素酶相比,总黄酮得率增加了54.93%。
In order to study the factors affected the yield of total flavonoids extracted from leaves of Ginkgo biloba by using cellulase-assistant extraction (EAE) technology, the single factor experiments were conducted, in which the effect of particle size, extraction time and cellulase amount on the extraction was studied respectively. Based on the experiments above, the particle size, extraction time and cellulase amount used were selected as the factors for the orthogonal experiments to find out the optimum operating conditions for the EAE of total flavonoids extracted from G. biloba leaves. The results of orthogonal experiments showed that the optimum operating conditions were as follows:the extracted particle size used was between 150 - 180 μm, the added amount of cellulase was 30 FPIU (Filter Paper International Units)/g( Ginkgo biloba leaves residue), and the cellulase extraction time was 50 rain. Using the mentioned optimum conditions, the total flavonoids extracted was 2.51% and increased by 54.93%, compared with the non-using the cellulase-assistant technology.
