摘要 目的:探讨白藜芦醇诱导人胃癌SGC7901细胞凋亡的作用。方法:以Res(10、20、40μg/mL)作用人胃癌SGC7901细胞,同时设DMSo和培养液作用为对照组。采用MTT法检测Res对人胃癌SGC7901细胞的生长抑制情况.荧光染色法观察细胞形态变化。琼脂糖凝胶电泳法分析SGC7901细胞的DNA片断化。结果:Res对SGC7901细胞生长有明显抑制作用,且随着剂量的增高(10~40μg/mL)而增高,最大抑制率达(53.39±5.32)%;荧光染色结果发现Res48h凋亡细胞增多,镜下可见较为典型的凋亡形态学变化:细胞变小,核皱缩,荧光强度增强.呈囊月或环状.核仁消失,新月体样改变,凋亡小体形成等;琼脂糖电泳出现明显的“梯状”条带,而对照组未出现梯状务带。结论;Res能押制人胃癌SGC7901细胞的生长并诱导其凋亡。 Objective:To investigate the effects of Resveratrol on the induction of apoptosis of human gastric cancer SGC7901 cells. Methods:Resveratrol(10,20 and 40μg/mL)affected human gastric cancer SGC7901 ceil line,and DMSO and culture liquid were used in control group. The growth inhibition rate of SGC7901 cells was detected by MTT method,the cell shape change was observed by discrepancy microscope and the caspase-3 activity was assssed by eolorimetry and the cell cycle was detected by flow cytometry (FCM). Results : Resveratrol inhibited the growth with the dose increase (10-40μg/mL)and the most inhibition rate was (53. 39±5.32) %. The suspend cells were increased,and the cell body,shrinked, rounded and smashed, was observed under discrepancy microscope. Resveratrol could increased the caspase-3 activities of SGC7901 cells in a dose-and time-dependent manner. Flow cytometry revealed that Res could decrease reproductive activity by inhibiting cell division. Conclusion: Resveratrol can inhibit human gastric cancer SGC7901 cell and its mechanism might act on Caspase-3, inducing apoptosis because of the increased suspend cell,the shrinked, rounded and smashed cell body under discrepancy microscope, therefore the cycle of tumor cell was affected and tumor cell was inhibited.
