摘要
对张宏达系统山茶属茶组植物19种和变种,应用ISSR标记对其进行亲缘关系的分析。从100个ISSR引物中,筛选得到11个多态性较好的引物,ISSR反应共得到166条可重复条带,多态性条带有155条,多态位点百分率(PPB)是93.4%,分析揭示茶组植物的Nei’氏遗传距离(He)是0.2951,Shanon多样性指数是0.4520;基于Jaccard相似性系数的UPGMA树型图表明,茶组植物明显地分为2个主要的类群("三室类群"和"五室类群"),ISSR表型特征的主成分分析(PCA)也支持了聚类分析结果。指出与普洱茶C.assam ica进行种间杂交育种,应优先选择茶组植物中C.irrawadiensis、C.dehungensis和C.taliensis。
The genetic relationship of 19 species and varieties in section Thea genus Camellia were analysed with ISSR technique. A total of 155 polymorphic bands out of 166 reproducible products were amplified from the selected 11 primers. Percentage of polymorphic loci (P) of Section Thea was 93.4 %. The genetic diversity within section Thea calculated from Nei's genetic diversity (He) was estimated to be 0. 2951 ,while Shannon indices (Ho) were 0. 4520;Cluster analysis of the Jaecard coefficient matrix using UPGMA method revealed that Section Thea were clustered into 5-locule ovary group and 3-locule ovary mainly, which was also lent support from the spatial representation of principle components analysis (PCA) of ISSR phenetic patterns. To cross-breeding new cultivars with C. assamica, C. irrawadiensis, C. dehungensis and C. taliensis were priority species among Sect. Thea.
