利用基因芯片分析非洲菊花序发育相关基因的表达 被引量:4

摘要 通过GenBank搜索已发布的花序发育基因序列,设计特异性的引物,采用PCR法扩增,获得了50个相应的基因片段,利用这些基因片段制备基因芯片。应用此芯片与已标记的非洲菊cDNA杂交,比较分析了同一基因在非洲菊不同发育时期的表达情况和同一基因在不同肥料条件下的表达情况。选取基因NM118013、AJ009726和AJ554702分别进行了RT-PCR验证,结果与芯片检测一致。本研究揭示了非洲菊花序基因时空和营养因子作用下的表达规律,有助于了解非洲菊花序的生长发育机理,进而指导非洲菊的遗传育种。 By using data from GenBank,capitulum development genes including MADS-box genes, NAC genes,LEAFY genes and ESTs were selected. The specific primers for capitulum development genes were designed, and the specific fragments amplified by PCR were cloned. The gene chip was made with these genes. Gene chip has been hybridized with tagged cDNA to compare the same gene expression in different growth stages and to analysis of the same gene expression under different fertilizer. At last NMllSO13 ,AJ009726 and AJ554702 were tested by RT-PCR, and the results were accord with chip examination. This research showed gene expression in different Gerbera capitulum and under different fertilizer by gene chip. The results were useful to understand development of C, erbera flower and guide its genetic breeding.
