摘要
对产木聚糖酶的毕赤酵母进行5L罐发酵研究,确定了5L罐发酵的最佳种龄是24h,最佳接种量是10%。通过对pH值、搅拌转速、温度、空气流量进行正交设计试验,得出结论:pH值和温度是影响产酶的主要影响因子,并结合实际情况,得到5L罐发酵产木聚糖酶的工艺条件:pH值为5.5,温度为30.0℃,空气流量3L/min,搅拌转速为300r/min,发酵130h,毕赤酵母发酵产木聚糖酶的酶活为2780U/mL。
The xylanase production by Pichia pastoris was studied in 5 L fermentor, by which the optimal cell age and inoculum were confirmed as 24 h and 10%, respectively. The pH, stirring speed, temperature and air flow rate during fermenation were investigated by orthogonal test. The results showed that the pH and temperature were major factors affecting xylanase production. The optimum parameters of xylanase fermentation in 5L fermentor were: pH 5.5, temperature 30.0℃, air flow rate 3L/min, stirring speed 300 r/min and fermentation time 130 h. Under above conditions, the e xylanase activity reached 2780 U/ml.
