血管活性肠肽对脑缺血再灌注损伤大鼠E-选择素水平的影响 被引量:1

Influence of vasoactive intestinal peptide on E-selectin in rat with focal cerebral ischemia-reperfusion

摘要 目的利用活体实验观察血管活性肠肽(VIP)对脑缺血再灌注后E-选择素(E-selectin)动态变化的影响,探讨VIP的神经保护作用。方法大鼠侧脑室注射VIP后,用线栓法制作大鼠大脑中动脉再灌注(MCAO)模型。通过神经学的评价对MCAO大鼠进行判定和筛选;选取再灌注后4个时间点,采用原位杂交法检测E-selectin mRNA的表达;应用免疫组化技术检测E-selectin的蛋白表达。结果VIP注射后E-selectin mRNA和蛋白在脑缺血再灌注各时间点缺血侧皮层和纹状体较对照组表达下降(P均<0.05)。结论VIP脑内注射可明显降低脑缺血再灌注后缺血区E-selectin mRNA和蛋白的表达,从而减轻再灌注损伤程度,提示VIP具有体外神经保护作用。 Objective It is to observe the influence of vasoactive intestinal peptide (VIP) on dynamic changes of E-selectin after focal cerebral ischemia reperfusion (CIR) in vivo and approach the neuroprotective effects of VIP. Methods After VIP was given by intracerebroventriclar injection, middle cerebral artery occlusion (MCAO) model was made by thread - tie method. The evaluation and screening of rats with successful occlusion of MCAO were performed by neurological testing. The successful model rats were randomly divided into four time point reperfusion respectively. The mRNA expression of E - se- lectin was detected by in situ hybridization method; and the protein expressive of E- selectin was detected by immunohisto- chemisty technique. Results Compared to the control groups, the mRNA and protein expression of E- selectin are remark- ablely down regulated in cerebral ischemic cortex and corpus striatum at four time point reperfusion after VIP injection (P 〈 0.05). Conclusion VIP injection can reduce the mRNA and protein expression of E- selectin after CIR and decrease the de- gree of CIR injury. It prompt that VIP has neuroprotective effect.
