摘要
为寻找治疗缺血性心脏病的新方法,我们采用DEAE-52离子交换柱层析盐梯度洗脱等方法首次从人胚胎心室肌中提取一种在体外检测中具有生物活性的蛋白类物质,即心肌生长因子,并对其纯度、氨基酸组成、N末端部分氨基酸序列及其同源性进行了研究.实验结果表明:该物质系高纯度的蛋白类物质,分子最3.7万道尔顿,含有17种氨基酸,N末端氨基酸序列为NH-Ser-GLy-Cys-Cys-Pro……,经蛋白文库同源性扫描,未发现同源物质,其临床意义正在进一步的研究之中.
We have first recovered and purified a protein material-myocardium Growth Factor (MGF) that has bioactivity in vitro from ventricle musule of human embryo with the methods of DEAE-52 cellulosion exchange chromatography, SDS-PAGE and so on for searching a new way to treat ischemic heart disease, and it has been studied ahout its purity (SDS-PAGE, carpillary zone electrphoresis), amino acid proximate analysis ,N-terminal amino acid sequence of this protein and its autoploidy analysis. Our results comfirm that MGF is high purified protein, its molecular weight is about 37000 Dalton, it contains 17 kinds of amino acid, N-terminal amino acid sequence is N-Ser-Gly-Cys-Cys-Pro. . . , the homogeneous material has been not recovered by scanning of protein libary. Its clinical manifications are to be studied by us.
